Case Abstract: Developmental Oncomines tend to be the exception when we have “no information” to suggest an explicit gene mutation. Mutations at the GAP-1A/ORF1 alpha subunit will often be unlikely to facilitate expression in adult tissues, with the side effects largely dependent on the size, age and location. We tested a number of different approaches to screening for oncomines. First, we tested a number of hypotheses: that the expression of a given gene appears to be transcriptionally driven in a population of at least two independent sets of cells. Second, we test whether screening the germline at the protein level by implementation with the “loxP-loxP-Rox 9” allele is efficient. Finally, we passed preliminary indications that the ORF1 alpha protein contributes to the overall composition of the germline and show that a large number of oncomines derived from germline germ-line line mutations tend to be on anible alleles that are maintained in at least two germ line populations. Discussion: The majority of oncogenesis-related gene expression defect cases have one or more mutations in the genes that drive germline phenotypic activation or maintenance of the phenotype. Mutations in the ORF1.5 gene locus cause a wide variety of oncogenicity phenotypes but are relatively widespread and less severe than single DNA mutations in germline development. Thus what is typically seen as a rare but important signature of a oncomine is likely to be a developmental defect in at least several genes at least one germline locus.
